1993-ban Shakur megalapította a „Thug Life” nevű együttest, legjobb barátaival, Big Syke-kal, Macadoshis-szal, mostohatestvérével Mopreme Shakurral és Rated R-rel. Az első és egyetlen albumuk, a „Thug Life Vol. 1”, 1994. szeptember 26-án jelent meg. A „Thug” szónak kettős jelentése van: egyrészt mozaikszóként a „The Hate U Gave Little Infants Fucks Everybody” kezdőbetűiből áll össze, másrészt Tupac az olyan hátrányos helyzetűekre is használta, akik mégis sokra vitték az életben. Ő maga is ilyen volt.
